Cellstripper™ Solution

Cellstripper is a non-enzymatic cell dissociation solution formulated with a proprietary mixture of chelators that gently dislodge adherent cells in tissue culture. It is an alternative to trypsin when used with either serum-free or serum-supplemented media. Because Cellstripper is a gentler reagent than trypsin, cells can be exposed to Cellstripper for longer periods of time. Cellstripper achieves superior results in comparison to enzymatic dissociation products.

Trace Elements

Trace Elements A provides copper, zinc, iron and selenium. Selenium aids in the detoxification of free radicals as a cofactor for GSH synthetase, while iron, copper, and zinc may be bound by serum protein. Trypan Blue Solution

Trypan blue is the most common stain used in cell count and viability assays. Viable cells exclude the dye, while nonviable cells absorb the dye and appear blue. Cells should be in suspension as single cells in buffered saline before counting. Trypan blue can be used in assays providing total cell count, viable cell count or nonviable cell count alone. This solution is provided at the common concentration of 0.4% (w/v).

Miller’s LB Broth

Miller’s LB broth is based on the original Lysogeny Broth (LB) media formulation developed by Luria and Bertani, which was designed for the rapid propagation of recombinant E. coli strains. Miller’s LB contains a 20-fold concentration sodium chloride compared to the original formulation, and is widely used for the growth and maintenance of cells in work with plasmid DNA and recombinant protein production.

Ham’s F-12 media was originally formulated for the serum-free culture of CHO cells, and is based on Ham’s F-10 medium with increased concentrations of choline, inositol, putrescine, and several amino acids. This medium is also suitable for carcinoma cells, rat skeletal myoblasts, Chinese hamster lung cells, and rat, rabbit, and chicken embryos.

Iscove’s Modification of DMEM

Iscove’s Modification of DMEM (IMDM) is a highly enriched synthetic medium optimized for rapidly proliferation cultures of high cell density. A modified form of Dulbecco’s Modified Eagle Medium (DMEM), Iscove’s contains additional amino acids, vitamins, selenium and HEPES buffer, and is formulated with potassium nitrate in place of ferric nitrate. The addition of albumin and transferrin may be required for certain cell types.

Leibovitz’s L-15 (Mod.)

Leibovitz’s L15 medium was originally developed without a sodium bicaronate buffer system for use in non-CO2 equilibrated environments. It is buffered by phosphates and the amino acid L-arginine, along with L-histidine and L-cysteine. The formation of acid metabolite byproducts is prevented by replacing glucose with galactose and sodium pyruvate. Examples of usage include the propagation of HEP-2, monkey kidney cells and primary explants of embryonic and adult human tissue. Leibovitz’s L-15 supports established cell lines as well as primary explants of embryonic and adult human tissue.

McCoy’s 5A medium was originally formulated to meet the culture requirements of Novikoff Hepatoma cells, and has been found to support the indefinite proliferation of Walker 256 carcinoma cells along with a wide range of human and rat normal and transformed cells. Originally modified by Iwakata and Grace, this version of McCoy’s 5A includes increased levels of inositol and glucose and contains L-glutamine.

Minimum Essential Medium (MEM)

Minimum Essential Medium (MEM) is a common cell culture medium developed from early work using Basal Medium Eagle (BME) with normal mammalian fibroblasts and certain subtypes of HeLa cells. This research indicated that additions made to BME aided cell propagation. MEM is modified with higher concentrations of amino acids to more closely approximate the protein composition of mammalian cells. When supplemented with serum, MEM has been used for cultivation of a wide variety of cells grown in monolayers, such as fibroblasts.

RPMI 1640

Named for the site at which it was developed, the Roswell Park Memorial Institute, RPMI 1640 is a modification of McCoy’s 5A containing an increased level of inositol. Examples of cell types for which this medium is appropriate include mammalian myeloma and hybridoma cells such as mouse hybridomas, human leukocytes, and B and T lymphocytes. RPMI 1640 is also used for suspension cultures or monolayer human leukemic cells.
